ABSTRACT

Objective:To formulate surgical strategies and guide the implementation of laparoscopic anatomical hepatectomy with preoperative simulative resection.Methods:Twenty-two cases of hepatocellular carcinoma undergoing laparoscopic lobe, segment, subsegment and combined segment liver resection following preoperative simulative resection from Sep 2020 to Jan 2022 were enrolled in this study retrospectively. We observed and analyzed the operation time, intraoperative blood loss, postoperative hospital stay and postoperative complication. Results:All patients underwent laparoscopic hepatectomy successfully according to the preoperative simulative resection plan without conversion, some of them adjusted plan according to preoperative simulative resection. The median operation time was 170.0 min, the median intraoperative blood loss was 150.0 ml, the median times of pringle maneuver was done on 4 episodes, and the median postoperative hospital stay was 6.5 days. There were no severe postoperative complications in all cases. Conclusion:Preoperative simulative resection can plan the range of surgical resection accurately by visualizing important anatomical structures, greatly helping the actual surgical hepatectomy.

ABSTRACT

OBJECTIVE@#To investigate the effect of rapamycin on Parkinson's disease (PD) and its underlying mechanism in mice.@*METHODS@#Sixty SPF adult male C57BL/6 mice were randomly divided into control group, model group and treatment group. 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine(MPTP) was used to induce Parkinson's disease in model group and treatment group. All mice were trained to cross the runway and were subjected to computer-assisted CatWalk. The numbers of tyrosine hydroxylase positive (TH) neurons in the substantia nigra (SN) were assessed by unbiased stereology using the optical fractionator method; protein expression was detected by Western blot analysis; and glutathione peroxidase (GSH-Px), malondialdehyde (MDA) and superoxide dismutase (SOD) were detected by spectrophotometry.@*RESULTS@#In the model group, a decrease in stride rate and an increase in variation of stance and swing were noted by CatWalk system (<0.05 or <0.01); the numbers of TH neurons decreased (<0.01); expression of p-Akt, p-S6K, p-S6 and p-ULK increased (all <0.01); LC3-Ⅱ/Ⅰ ratio decreased (<0.01); MDA level was elevated while the levels of SOD and GSH-PX were reduced (all <0.01). Compared with the model group, after treated with rapamycin, the abnormal behavior including the stride length, variation of stance and swing and step patterns induced by MPTP were all improved (<0.05 or <0.01); the numbers of TH neurons increased (<0.05); the expression of p-Akt, p-S6K, p-S6 and p-ULK was suppressed (all <0.01); the LC3-Ⅱ/Ⅰ ratio was upregulated (<0.05); MDA level decreased while the levels of GSH-Px and SOD increased (all <0.01).@*CONCLUSIONS@#Rapamycin inhibits the activation of mTOR pathway, which contributes to protect against the loss of dopaminergic neurons and provide behavioral improvements in mice with Parkinson's disease. These results are partially related to the ability of rapamycin in inducing autophagy and reducing oxidative stress.

ABSTRACT

In this work, retinal penetration of fluorescein was achieved in vitro by covalent attachment of taurine to fluorescein, yielding the F-Tau conjugate. Nuclear magnetic resonance (NMR) and high resolution mass spectrometry (HRMS) were used to confirm the successful synthesis of F-Tau. The cellular uptake of F-Tau in adult retinal pigment epithelial cells (ARPE-19) and human retinal microvascular endothelial cells (hRMECs) was visualized via confocal scanning microscopy. The results indicated an improvement of solubility and a reduction of logP of F-Tau compared with fluorescein. As compared with fluorescein, F-Tau showed little toxicity, and was retained longer by cells in uptake experiments. F-Tau also displayed higher transepithelial permeabilities than fluorescein in ARPE-19 and hRMECs monolayer cells (P<0.05). These results showed that taurine may be a useful ligand for targeting small-molecule hydrophobic pharmaceuticals into the retina.

ABSTRACT

OBJECTIVE To investigate the influence of tea polyphenol in alcoholic liver(hurted) rat by observing its blood plasma endotoxin level, and to provide the direction of new(treatments) and means for the prevention of alcoholic liver disease.METHODS Thirty S-D rats(15 female,15 male) were divided into five groups randomly(noted as A,B,C,D,and E,with equal male and female),and fed in the cleanliness class laboratory.Excepting group E which was given saline, all other groups were given alcohol(56% Hongxing spirits) with the dosage of 7 g/kg once everyday. At the same time, rats in groups B,C and D were given tea polyphenol with the dosage of 0.05,0.125,0.25 g/kg.In the end of the fourth week, got the rats vein blood to determinae serum ALT,AST level and blood plasma endotoxin level.RESULTS The serum AST of the group A was higher than other 4 groups;the blood plasma endotoxin level in groups B,C,and D were(consequently) lower than in group A.CONCLUSIONS The tea polyphenol could reduce the blood plasma endotoxin level in alcoholic liver hurted rat.Its possible mechanism is to repress the isotopy of bowel flora, and to maintain the normal function of bowel mucous membrane. This research provides a new way for curing the alcohol liver hurt.
